Vinegar is an acid and will react with calcium sand such as aragonite. The composition of the sand has very little to do with whether or not it can be used. As long as the sand is free of impurities, silica is no worse than aragonite.

The cancer is silicosis and it comes from breathing in silica dust. As long as the sand is underwater the risk of cancer from the silica dust shouldn't be a problem.
