Unhappy ---help-me---

I get a trouble in my job. There is a word I don't know in a article I am reading."Preplate the cells for 15min on a collagen coated dish before moving the cells remaining in suspension to a new collagen coated dish". What is"Preplate" meaning? Thank you very much.

Maybe you mean palpate - which means to examine by touch especially medically

Preplate technique allows the seperation of cells based on their differences in adhesion characteristics.


Preplate is a technique and the media used for adherence might vary with which cells one would like to seperate.

It means this:

1. Add x concentration of y volume collagen to plate for 15 min. Wash it off with preferred solution (DPBS or media without additives - it eliminates the acidicity of collagen) or dry completely.
2. Remove an aliquot of the cells to said plate. Ususally it is trypsinized first. Primary myoblasts grow as adherant cells, not cells in suspension.
3. Incubate with myoblast media. It can contain serum or not. Check the paper to make sure. I have used both situations. It depends on your media components.

That is true, but I believe that in this case all he needs to do is coat the plate with collagen to make sure that myoblasts stick to the tissue culture plate.

Preplating with certain substances can preferentially remove or retain cells of interest. I use this technique to a certain degree to isolate retinal ganglion cells (from the eye) to get rid of contaminating macrophages. The macrophages stick to any surface within 20 minutes while my RGCs stay in suspension during this time. It allows me to further purify my RGCs.
