I think you need to put the anenome in the tank...
I think you need to put the anenome in the tank first....The stress from moving is tough on it then to add a clown will be even more stressful...Clowns can really beat them up....

I dont really think there is a best way to...
I dont really think there is a best way to aquascape but I would probably try to use smaller rocks to build your formations, that way you can control the spread better...Larger rocks make that...

It depends on where you are wanting to get most...
It depends on where you are wanting to get most of your flow from....If you want to just get the water back from your sump to your tank and let a closed loop, tunze, etc do the rest then I wouldnt do...
